摘要
目的探讨人突变载脂蛋白E4(apolipoprotein E4,ApoF4)基因表达对转基因鼠血脂代谢的影响。方法应用显微注射法建立人突变ApoE4基因的转基因鼠,分子杂交鉴定。酶法测定人突变ApoE在F1代小鼠染色体上的整合与在血中的表达。酶法测定小鼠血清三酰甘油(triglyceride,TG)和血清胆固醇(cholesterol,TC)水平。结果人突变ApoE4基因稳定地整和于F1代小鼠染色体上并高效表达于血清中。12月龄转基因鼠血脂代谢[血清TG水平(2.89±0.31)mmol/L,血清TC水平(3.02±0.14)mmol/L]与同龄对照组鼠[血清TG水平(2.05±0.12)mmol/L,血清TC水平(2.11±0.01)mmol/L]比较差异有统计学意义(P<0.05,P<0.01)。结论人突变ApoE4基因的异常表达影响到了小鼠的血脂代谢。
Objective To study the influences of the mutants of human apolipoprotein E4 (ApoE4) gene on the lipoprotein metabolism in transgenic mice. Methods The human ApoE4 transgenic mice were generated via mieroinjection. The hybridization and ELISA techniques were used for identification of integration and expression of human ApoE4 gene at the mice chromosome and in the serum of F1 transgenic mice. The serum total cholesterol (TC) and serum triglyeeride (TG) were determined with enzymatic procedure. Results In F1 transgenie mice the h- ApoE4 gene integrated at the chromosome and expressed in serum stably. The level of lipoprotein metabolism were high in transgenic mice of 12 - month age [ TG, (2.89 ±0.31 ) mmol/L, TC, (3.02± 0.14) mmol/L] than that in the respective control mice of 12 - month age [ TG , (2.05 ± 0.12)mmol/L,TC, (2.11 ±0.01 ) mmol/L] ( P 〈 0.05, P 〈 0.01 ) . Conclusion The overexpression of apoE gene possibly impairs the lipoprotein metabolism.
