基于动物热活性检测的肾阴虚/肾阳虚模型寒热属性差异研究 被引量:19

Investigation on Cold /Heat Characters of Kidney Yin /Yang Deficiency Models by Zoogenic Thermal Activity

摘要 目的:从动物热活性角度考察中医肾阴/阳虚体质动物模型的科学性与可靠性。方法:采用sc氢化可的松50 mg.kg-1×5 d或25 mg.kg-1×10 d分别复制小鼠肾阴虚/肾阳虚模型,采用冷热板示差法记录动物在不同温区分布比例,同时观察动物采食量、饮水量、呼吸耗氧量、三磷酸腺苷酶(ATPase)、超氧化物歧化酶(SOD)活性及环磷酸腺苷/环磷酸鸟苷(cAMP/cGMP)等指标,以考察动物体质差异与自身能量代谢及获取环境能量补偿行为的关系。结果:肾阴虚动物在冷热板冷温区停留比例造模前5 d显著提高(P<0.05),后5 d逐渐恢复正常,机体能量代谢明显提高(P<0.05);肾阳虚证体质模型动物在冷热板高温区停留比例前3 d有所下降,后7 d显著提高(P<0.05或P<0.01)、机体能量代谢明显降低(P<0.05),结果与以往报道一致。结论:该文建立的冷热板示差法可以客观地表证肾阴/阳虚体质模型动物间的差异,为中医证候动物模型研究提供了新的方法,也为中药寒热药性的评价提供新的理论与技术支持。 Objective: To investigate the objectivity and validity of animal model for deficiency of kidney yin /yang in traditional Chinese medicine by means of zoogenic thermal activity.Method: Deficiency of kidney yin model and deficiency of kidney yang model in mice were duplicated by subcutaneous injection of hydrocortisone at the dose of 50 mg.kg-1 for 5 d or 25 mg.kg-1 for 10 d,respectively.The remaining rate of animals in different temperature zone were recorded by the hot cold plate instrument;simultaneously,the drink and food taking,the oxygen consumption and activities of adenosine triphosphatase(ATPase) and SOD,and cAMP /cGMP were detected,so as to investigate the relationship of physical status with energy metabolism /compensation from environment.Result: The remaining rate in hot plate of kidney-yin deficiency model was decreased notably(P 0.05) in the first 5 days,and then return to normal condition in the last 5 days,and their capability of energy metabolism was decreased significantly(P 0.05);while the remaining rate in hot plate of kidney-yang deficiency model mice was decreased in the first 3 days and then increased significantly(P 0.05 or P 0.01),and their capability of energy metabolism was increased significantly(P 0.05).The result was consistent with the previous reports in literatures.Conclusion: The method of animal differential distribution on hold cold plate could show the difference among cold /heat physical model animals,could be used as a novel mean for evaluation animal models of symptoms in traditional Chinese medicine and the nature of traditional Chinese drugs.
