摘要
以3-乙酰基-2-氯吲哚为原料,在丙酮的碱性溶液中发生烷基化和苄基化反应对氨基进行保护。探讨了反应温度及溶剂对反应产率的影响,在优化的反应条件下以良好到优异的产率(78.5%~97.7%)合成了N-烷基或苄基取代-3-乙酰基-2-氯吲哚。它们的结构通过红外光谱、核磁共振氢谱、质谱以及元素分析得以证实。
3-Acetyl-2-chloro-indole(1)was used as the starting material to perform N-alkylation and N-benzylation reactions in a basic acetone solution to give N-alkyl or benzyl substituted-3-acetyl-2-chloro-indoles.The effects of reaction temperature and solvent on the product yields were investigated,and the products N-substituted-3-acetyl-2-chloro-indoles were obtained in good yields(78.5%~97.7%)under optimal conditions.The structures of the target compounds were confirmed by IR,1HNMR,MS and elemental analysis.
