Expression of mBD-4 during nervous system development process of mouse embryo

摘要 目的观察β-防御素4(mouseβdefensin 4,mBD-4)在小鼠胚胎神经系统发育不同时期的基因表达及脂多糖(lipopolysaccharide,LPS)诱导孕鼠后mBD-4在胚鼠神经系统的的表达情况。方法孕鼠分为LPS组(n=10)和正常对照组(n=10),取不同时期(5.5、8.5、11.5、17.5天)小鼠胚胎,LPS组于胚胎提取1天前母鼠腹腔注射LPS,正常对照组同时间注射同量的生理盐水,胚胎取出后石蜡包埋切片,免疫组化方法和RT-PCR技术检测mBD-4的阳性表达。结果 mBD-4在小鼠胚胎早期第5.5天的神经组织中未见固有表达,到胚胎第8.5天开始有少量固有表达,到胚胎中晚期,固有表达含量增多;LPS组中胚胎神经组织中自5.5天有阳性表达,且随着胚胎的发育其阳性表达越强。结论①mBD-4在小鼠胚胎神经系统中固有表达,在胚胎第8.5天有少量表达,且随着神经系统的发育成熟,mBD-4在其神经组织中表达含量越多。③mBD-4在LPS的诱导下,其在神经组织中表达明显增高。 Objective To observe the expression of beta-defensin 4(mouse beta defensin 4,mBD-4) at the different stages of the nervous system development of mouse embryonic and that if the pregnant mice were induced by lipopolysaccharide(LPS).Methods The mouse embryonic were extracted at different time(5.5,8.5,11.5and 17.5 days),the pregnant mice were divided into LPS group(n=10) and normal control group(n=10),the pregnant mice of LPS group were intrapertinally injected into LPS and the normal control group at the same time of the injection with normal saline,embryo take out after paraffin-embedded chip,immunohistopathologic method and RT-PCR to measure the positive expression of hBD-4.Results mBD-4 have no expression in 5.5 mouse gestation day,and then it began to have a small amount of constitutive expression in the 8.5 mouse gestation day,following the embryo of advanced,increase natural expression were observed;the embryos in LPS group have a positive expression in 5.5 gestation day,following the development of embryo,the positive expression were more and more strong.Conclusion mBD-4 was constitutive expressed in mouse embryonic cyclophos phamide,until 8.5 gestation day,and with the nervous system development and maturation,mBD-4 expression was increase.mBD-4 was increase expression level in the cyclophos phamide by LPS induced.
