摘要
目的研究足叶乙苷长循环脂质体的小鼠体内代谢动力学。方法采用HPLC测定血药浓度,考察脂质体在小鼠体内的代谢动力学行为。结果足叶乙苷线性范围为10.5~210μg.mL-1,回收率在90%以上,精密度符合要求。长循环脂质体的消除半衰期t1/2和AUC分别是注射剂的17和3.5倍。结论足叶乙苷长循环脂质体在小鼠体内的消除半衰期较长,能达到长循环目的。
OBJECTIVE To investigate in vivo pharmacokinetics of etoposide long-circulating liposomes in mice.METHODS Plasma concentration was determined by a validated HPLC assay.In vivo pharmacokinetic dynamic behavior of liposomes was analyzed.RESULTS The linear range of etoposide was 10.5-210 μg·mL^-1,recovery ratio was between 90% and 100%,and accuracy met the requirements.The elimination half-life and AUC of long-circulating liposomes were 17 and 3.5 times as injection preparations.CONCLUSION Etoposide long-circulating liposomes had long half-life of elimination,can achieve long-circulating profile.
