摘要
目的:采用比较蛋白质组学方法分析正常与神经源性休克死亡蛋白表达差异。方法:外力打击兔颈动脉窦建立神经源性休克死亡模型,提取死亡组与对照组左心室肌蛋白进行二维电泳和凝胶图像分析,对差异蛋白质用HPLC-Chip/Ms纳流液质联用技术进行序列分析,经Spectrum Mill搜索UniProtKB/SWISS-PORT后鉴定蛋白质。结果:筛选出3个神经源性休克死亡组表达上调的蛋白质:封闭蛋白1、过氧化物酶增殖物激活受体δ、角蛋白15,1个表达下调的蛋白质甲基转移酶4。结论:神经源性休克发生了心肌蛋白表达改变,鉴定出的4种蛋白是潜在的神经源性休克生物标记物,可成为神经源性休克死亡诊断的客观依据。
Objective:Analysis of proteins expressed differentially between neurogenic shock death group and normal control group by comparative proteomics analysis.Methods:To establish neurogenic shock death model by hitting on the carotid sinus in rabbits.To extract proteins from the left ventricle of death group and normal control group.After 2-dimensional electrophoresis and PDQuest for gel image analysis,the MS / MS of the peptides from the differential proteins digested by tripsin were analyzed by HPLC-Chip/MS and the original data were pre-processed by Spectrum Mill then searched in UniProtKB/SWISS-PORT database.Results 3 proteins upregulated significantly in established neurogenic shock death group were identified,including Claudin I.Peroxisome proliferator activated receptor delta.Keratin 15,and as well as 1 proteins downregulated significantly including methyltransferase-like protein4.Conclusion:The proteins expression have changed in neurogenic shock death group.The proteins which were identified are latent biomarkers and can offer possible diagnosticate datas of neurogenic shock death.
