摘要
目的:研究PI3K抑制剂LY294002对SGC7901胃癌细胞增殖凋亡及相关调控因子的影响。方法:MTT法测定细胞增殖,流式细胞术AnnexinV-FITC/PI染色法检测细胞凋亡率,RT-PCR检测NAG-1、Skp2mRNA的表达情况。结果:LY294002可抑制SGC7901的生长,呈时间-剂量依赖性,细胞表现出形态学改变;经10、30、50μmo.lL-1的LY294002作用12h的凋亡率依次为(12.7±0.42)%、(36.5±0.45)%、(64.1±0.79)%,明显高于空白对照组的(2.3±0.36)%(均P<0.01);LY294002可上调NAG-1mRNA表达,下调Skp2mRNA表达。结论:LY294002可改变胃癌细胞株SGC7901Skp2和NAG-1基因的表达,抑制细胞增殖,诱导细胞凋亡,其机制可能与其调控NAG-1、Skp2mRNA表达水平有关。
Objective:To explore the influence of PI3K inhibition by LY294002 on the proliferation and correlated regulatory factors of gastric carcinoma cell SGC7901.Methods:The effect of LY294002 on the proliferation of gastric cancer cell were evaluated by MTT assays.Flow cytometry analysis was used to determine the apoptosis rate.RT-PCR was applied to measure the expression of NAG-1(Non-steroidal anti-inflammatory drug activated gene-1)and Skp2(S-phase kinase associated protein2)mRNA.Results:Administered with LY294002,the proliferation of gastric cancer cell decreased in dose-and time-dependent manners.The cell apoptosis rates after the treatment with 10,30,50 μmol·L^-1 LY294002 for 12 h were(12.7±0.42)%,(36.5±0.45)%,(64.1±0.79)% respectively,which were higher than those in control group(2.3±0.36)%(P〈0.01).The expression of Skp2 mRNA was down-regulated while that of NAG-1 mRNA was up-regulated.Conclusion:LY294002 could inhibit proliferation and induce apoptosis of SGC7901 cells,possibly through the regulation of Skp2 and NAG-1 expressions.
