摘要
目的:探讨白藜芦醇(Res)对裸鼠胃癌移植瘤组织中凋亡相关基因人程序化死亡分子5(PDCD5) mRNA表达的影响。方法:将BGC-823细胞复苏后,实验组采用200μmol/L的Res处理,同时设空白对照组(培养液对照组)、顺铂组(3mg/L)及联合用药组(Res 200μmol/L+顺铂3mg/L)。处理72h后将细胞种植于裸鼠皮下构建裸鼠胃癌移植瘤模型;采用原位杂交方法检测移植瘤组织细胞中PDCD5 mRNA的表达。结果:单用Res(FRes=1168.620,P<0.001)或顺铂(F顺铂=1283.560,P<0.001)均能增加裸鼠胃癌移植瘤组织中PDCD5 mRNA的表达,Res和顺铂有交互作用(F交互=72.150,P<0.001),2者联合应用后,裸鼠胃癌移植瘤组织中PDCD5 mRNA的表达增高更明显。结论:Res可能通过上调PDCD5的表达而发挥抑癌作用。
Aim:To explore the effect of resveratrol on mRNA expression of apoptosis-related gene,human programmed cell death 5(PDCD5),in gastric carcinoma tissue carried by nude mice.Methods:The human gastric adenocarcinoma BGC-823 cells were handled by 200 μmol/L resveratrol,and blank control group(medium control group),cisplatin(DDP) control group(3 mg/L),and combination group(200 μmol/L resveratrol + 3 mg/L DDP) were set up.72 h later,BGC-823 cells were injected into the skin of nude mice to establish transplanted tumor model.The PDCD5 mRNA in transplanted tumor tissue was examined by in situ hybridization.Results:Alone resveratrol(F = 1 168.620,P 0.001) or cisplatin(F = 1 283.560,P 0.001) could increase the mRNA expression of PDCD5 in transplanted tumor tissue.There was interaction(Finteraction = 72.150,P 0.001) between resveratrol and cisplatin,and the mRNA expression of PDCD5 in transplanted tumor tissue were much higher in combination group.Conclusion:The anti-tumor effect of resveratrol may realized by up-regulating the expression of PDCD5.
