摘要
通过分子生物学方法分离鉴定了4个新的梨SFBB-gamma基因,并对其序列进行了生物信息学分析。SFBB-gamma基因的特异性引物对4个梨品种进行PCR扩增结果表明,4个品种均扩增出一条1 250 bp左右的条带;通过DNA测序和生物信息学分析方法从‘鸭梨’、‘红皮酥’、‘中梨二号’和‘兰州软儿’等梨品种中分离鉴定了SFBB21(EU422961)、SFBB26(EU422961)、SFBB31(EU422959)、SFBBd-gamma(EU422962)基因。聚类分析中发现,仁果类SFBB基因和核果类和SFB基因明显的分成两类,仁果类中梨SFBB基因和苹果SFBB基因又各分成一类,这与物种的亲缘关系相符合。新鉴定的梨SFBB基因中从白梨中分离鉴定的SFBB21-gamma基因与梨其他SFBB-gamma基因的亲缘关系最远。该研究为梨自交不亲和机理研究和遗传改良提供理论基础。
SFBB-gamma gene of four Chinese pears have been identified with molecular biological methods.Specific primer of SFBB-gamma gene was used in PCR amplification of four Chinese pears,the results show that a common fragment of about 1 250 bp was generated in four cultivars;by means of DNA sequencing and bioinformatic analysis,the SFBB21(EU422961),SFBB26(EU422961),SFBB31(EU422959),SFBBd-gamma(EU422962) gene were obtained from 'Yali','Hongpisu','Zhonglierhao'and'Lanzhouruaner'.Cluster analysis of four pears shows that SFBB gene of kernel fruits and SFB gene of stone fruit can be divided into two kinds obviously;SFBB gene of pear and SFB gene of apple were further divided into two groups in kernel fruits,this result is consistent to the result of morphological classification.Among these new SFBB genes,the SFBB21-gamma gene in the Pyrus bretschneideri has the lowest genetic relationship with other SFBB genes.This study would provide theoretical basis for mechanism of pear self incompatibility and genetic improvement in pears.
