摘要
目的探讨缺氧诱导因子-1(hypoxia-inducible factor1,HIF-1)与血管内皮生长因子(vascular endothelial growth factor,VEGF)是否作为2个相关因子参与子宫内膜腺癌的发生、发展,以及其表达是否与雌激素调控有关。方法选择2008年5月~2009年10月手术切除的子宫内膜腺癌标本30例(内膜腺癌组),同期剖宫术后的非典型增生(非典型增生组)、子宫内膜单纯性增生(单纯增生组)及正常子宫内膜(正常子宫内膜组)标本各10例,检测HIF-1α与VEGF的表达,分析HIF-1α和VEGF之间的相关性及与雌激素受体(ER)的关系。结果 HIF-1α在内膜腺癌组、非典型增生组阳性表达率分别为96.7%、70.0%,显著高于单纯增生组和正常子宫内膜组的20.0%、20.0%,差异有统计学意义(P〈0.01);内膜腺癌组VEGF表达率为100%,显著高于非典型增生组及单纯性增生组、正常子宫内膜组的80.0%、70.0%、70.0%,差异亦有统计学意义(P〈0.01)。内膜腺癌组HIF-1α、VEGF均高表达,并呈正相关(r=0.315,P〈0.05);内膜腺癌组HIF-1α、VEGF与ER间为正相关关系(r=0.478,P〈0.01)。结论 HIF-1α、VEGF在子宫内膜腺癌的发生、发展中发挥作用,其表达水平受雌激素调节影响,HIF-1α可能通过上调其靶基因VEGF的蛋白表达,促进肿瘤组织血管的形成,加速肿瘤的发展和侵袭。
Objective To investigate correlations between hypoxia inducible factor 1 (HIF 1) and vascular endothelial growth factor (VEGF) and their involvement in the occurrence of endometrial adenocarcinoma and development,and to observe the relationship of their expression in endometrial adenocarcinoma and estrogen regulation. Methods 30 specimens of endometrial adenocarcinoma (endometrial adenocarcinoma group) resected during May 2008-October 2009 were selected; 10 of atypical hyperplasia after cesarean(atypical hyperplasia group),10 of simple hyperplasia(simple hyperplasia group),10 of normal endometrial tissue(normal endometrial group). The expression of HIF 1α and VEGF were detected and the relevance of HIF1α and VEGF was analyzed,as well as their relationship with estrogen receptor (ER). Results The positive expression rates of HIF1α in endometrial adenocarcinoma group and atypical hyperplasia group were 96.7% and 70.0% respectively,significantly higher than that of simple hyperplasia group(20%) and normal endometrial group(20%),The differences were statistically significant (P0.01); The positive expression rate of VEGF in endometrial adenocarcinoma group was 100%,significantly higher than that of atypical hyperplasia group(80.0%),simple hyperplasia group(70.0%) and normal endometrial group(70.0%),The differences were also statistically significant (P0.01). HIF1α and VEGF were both highly expressed in endometrial adenocarcinoma group,there was a positive correlation (r=0.315,P0.05),and they also had a positive correlation with ER (r=0.478,P0.01). Conclusion HIF 1α and VEGF can play a role in endometrial adenocarcinoma of the occurrence and development,and their expression is affected by estrogen regulation,HIF 1α may up-regulate the protein expression of its target genes (VEGF),so as to promote tumor blood vessels formation,accelerate tumor growth and invasion.
