摘要
目的研究姜黄素对鼻咽癌增殖和迁移的影响,探讨姜黄素抗鼻咽癌的可能机理。方法考马斯蓝染色和图像分析法检测不同浓度姜黄素(0、10、20、40μmol.L-1)对H5细胞骨架的影响;细胞刮痕实验观察不同浓度姜黄素对H5细胞迁移能力的影响;以免疫细胞化学方法检测EGFR、PCNA、PI3K、Tubulin-β在各组细胞中的表达情况;RT-PCR方法检测不同浓度姜黄素对H5细胞EGER mRNA表达的影响;通过体内H5裸鼠移植瘤实验,观察姜黄素对移植瘤生长的影响,免疫组织化学方法检测不同浓度姜黄素作用H5裸鼠移植瘤组织中EGFR、PCNA、PI3K、Tubulin-β的表达。结果随着姜黄素浓度的增加,刮痕边缘的H5细胞向划痕外迁出减少(P<0.05);H5细胞骨架染色逐渐降低(P<0.01);不同浓度的姜黄素可抑制H5细胞中EGFR、PCNA、PI3K和Tubulin-β的表达,并使EGFR mRNA表达下调。姜黄素抑制H5裸鼠移植瘤生长,用药组的移植瘤组织坏死更为广泛(P<0.01);高剂量姜黄素处理组H5裸鼠移植瘤组织中的EGFR表达与对照组相比明显减弱(P<0.05)。Spearman相关分析显示EGFR、PCNA和Tubulin-β随姜黄素剂量的增高表达减弱,呈剂量效应关系(P<0.05或0.01);EGFR与PI3K、EGFR与Tubulin-β表达具有相关性(P<0.05);PCNA与PI3K表达具有相关性(P<0.05)。结论姜黄素可能通过下调EGFR影响细胞骨架,从而抑制鼻咽癌细胞H5的增殖及迁移。
Objective To observe the effect of curcumin on the proliferation and migration of nasopharyngeal carcinoma and to explore the possible mechanism of anticancer activities of curcumin on nasopharyngeal carcinoma.Methods The cystoskeleton of H5 cells treated with curcumin(0,10,20,40μmol·L^-1) were evaluated by Coomassie blue staining and image analysis.Cell scrape experiment was performed to observe the capability of migration of H5.The expression of EGFR,PCNA,PI3K and Tubulin-β in each group were measured by immunohistochemistry.RT-PCR was used to detect EGFR mRNA of H5 cells treated with curcumin in different concentration.The effect of the different doses of cucumin on the growth of implanted tumors were observed in nude mice experiment of H5 in vivo.The expressions of EGFR,PCNA,PI3K and Tubulin-β in implanted tumors were examined by immunohistochemistry.Results With concentration of curcumin increasing,less H5 cells were outside the margin of the scrape(P〈0.05)and the cystoskeleton staining were significantly reduced(P〈0.01).The curcumin with different concentration inhibited the expression of EGFR,PCNA,PI3K and Tubulin-β,with a down-regulation of EGFR mRNA in H5 cells.Curcumin restrained the growth of H5 implanted tumors.Necrosis in xenografts of medication group was significantly more extensive than that in control group.The expression of EGFR in H5 implanted tumors treated with high dose curcumin were weakened significantly compared with the control group(P〈0.05).Spearman correlation analysis showed that the expression of EGFR,PCNA and Tubulin-β decreased in dose dependent manner with curcumin(P〈0.05 or 0.01).There were significant correlation between the expression of EGFR and Tubulin-β,EGFR and PI3K,also between PCNA and PI3K(P〈0.05).Conclusion Curcumin inhibited the proliferation and migration of nasopharyngeal carcinoma cell H5 through the way of influencing the cystoskeleton by the downregulation of EGFR.
