摘要
目的研究改良后的甘油长期冷冻保存法保存的角膜缘组织的超微结构及临床应用效果。方法取角膜移植术后的新鲜角膜缘组织及严重眼球裂伤后健康的角膜缘组织18例(18眼)以改良甘油长期冷冻保存法进行保存,保存时间3~24个月,其中12例应用于临床板层角膜缘移植术,6例保存时间大于半年的角膜缘组织进行光镜及电镜检查。结果11例(12眼)手术均顺利,术后随访6~24个月,无植片融解脱落,10眼植片透明,1眼植片呈半透明状,1眼植片部分血管化。光镜及电镜检查显示角膜组织的显微结构及超微结构得到良好保存。结论改良甘油长期冷冻保存的角膜缘组织具有一定活性,应用于角膜缘移植成功率高,能够满足临床应急需要。
Objective To study the ultrastructure of human corneal limbal tissues preserved by improved long-term eryopreservation with glycerol, and its clinical application. Method 18 human limbal tissues were remanded after PKP ( penetrating keratoplasty, PKP) or got from health limbal tissues of severe ocular laceration. Those limbal tissues were preserved by improved long-term cryopreservation with glycerol for 3-24 months. 12 limbal tissues were applied to LKP (lamellar keratoplasty, LKP) and 6 limbal tissues that were preserved for 6 months accepted optical microscope and electron microscope examination. Results All of the surgeries in the 11 cases (12 eyes) were successful, and the pa- tients were followed up fur 6 to 24 months. 10 grafts were transparent, 1 graft was translucent, and 1 graft was neovascularization. Optical microscope and TEM examination showed that the ultra- structure of corneal limbal tissue was well-preserved. Conclusion The improved long-term cryopreservation with glycerol could preserve some viability of human limbal tissues. It is a proper way of preservation that could be used for lamellar ke,'atoplasty in clinic when there have no fresh corneas.
