摘要 目的探讨灯盏花素抑制肾上腺素诱导的心肌细胞内钙升高的机制。方法培养乳鼠心肌细胞,经Fluo-3/AM染色后应用激光共聚焦技术检测细胞内钙水平。结果灯盏花素(10、30、100μM)显著抑制肾上腺素诱导的细胞内钙升高及细胞体积增大。灯盏花素抑制氯化钾(KCl)诱导的细胞外钙内流,但对L-型钙通道开放剂BAYK8644诱导的细胞内钙升高无影响。另外,灯盏花素能够抑制咖啡因介导的细胞内钙升高。结论灯盏花素通过抑制外钙内流和内钙释放降低细胞内游离钙的浓度,其抑制外钙内流的作用与L-型钙通道无关。 Objective To investigate the mechanism of scutellarin suppressed the increment of intracellular calcium induced by Epinephrine.Methods Neonatal cardiac myocytes were loaded with Fluo-3/AM,a calcium indicator.The intracellular calcium level was examined using laser scanning confocal microscope.Results Cultured neonatal cardiac myocytes exposed to Epinephrine appeared significant enlargement of cell size and the increment of intracellular calcium.Treatment with scutellarin significantly inhibited these alterations.Scutellarin apparently inhibited KCL induced increment of intracellular calcium,while produced no effect on L-type calcium channel opener,BAYK8644,caused intracellular calcium increment.Moreover,scutellarin pretreatment alleviated caffeine induced intracellular calcium release.Conclusion This study indicated scutellarin suppressed the intracellular increment by inhibiting both extracellular calcium influx,which is independent of L-type calcium channel,and intracellular release.
