摘要
目的检测经^(137)Csγ射线辐照红细胞悬液后淋巴细胞凋亡相关蛋白Fas/FasL、Bcl-2/Bax的表达水平,并分析其临床意义。方法利用Fas-异硫氰酸荧光素(FITC)、FasL-FITC、Bcl-2-FITC、Bax-FITC荧光标记抗体,应用单色流式细胞术检测辐照血液与未辐照血液的淋巴细胞凋亡相关蛋白的表达。结果经25 Gy剂量的^(137)Csγ射线辐照的红细胞悬液,淋巴细胞凋亡蛋白Fas、FasL表达显著升高(P<0.05),Fas/FasL的比值差异无统计学意义(P>0.05);而凋亡蛋白Bcl-2、Bax表达明显下降(P<0.05),且Bcl-2/Bax的比值下降更加显著(P<0.01)。结论 ^(137)Csγ射线辐照可促使离体血液淋巴细胞凋亡,并且可应用凋亡相关蛋白Fas、FasL、Bcl-2/Bax比值作为判断离体血液淋巴细胞经^(137)Csγ射线辐照后是否灭活的参考指标。
Objective To investigate the expressions of Fas/FasL.Bcl 2/Bax on the Lymphocyte of blood irradiated by 137Cs γ-ray. Methods We segregated the Lymphocytes of irradiated blood and normal controls by separating medium,then detected the expressions of apoptosis related proteins by flow cytometry(FCM). Results Compared with normal controls,the expressions of Fas and FasL on the Lymphocyte of blood irradiated by 25Gy 137 Csγ ray were improved significantly(P〈0. 05), but the odds of Fas/ FasL was of no significance(P〈0.05). Expressions of Bcl-2, Bax and the odds of Bel-2/Bax were decreased(P〈0.05), but the later was more notable(P〈0.01). Conclusion 137 Cs γ-ray irradiation can make the Lymphoeytes of blood in vitro apoptosis. Besides, Fas,FasL.Bcl-2/Bax maybe the reference makers judged whether the Lymphocytes of blood irradiated by 137Cs γ-ray in vitro was inactived or not.
