摘要
目的针对肿瘤生存环境进行研究,观察在肿瘤间质液(痰浊)影响下对体外培养的S180肿瘤细胞生长增殖的影响作用。方法取传3代S180肉瘤实体瘤,经研磨,用生理盐水与瘤重按照2:1比例稀释,以每只小鼠0.2mL右侧腋下接种混悬液,制备荷瘤鼠。至2周,肿瘤长至蚕豆大小。取瘤、研磨、离心、过滤,取不含肿瘤细胞的肿瘤间质液(痰浊)、间质液(痰浊)以3种不同质量浓度各20μL,对照组用生理盐水20μL,观察对S180肿瘤细胞的生长影响;MTT法观察肿瘤细胞增殖情况。结果提取的肿瘤间质液中不含肿瘤细胞,不同时间各组OD值差异有统计学意义,在时间上存在质量浓度递增关系;S180肿瘤细胞的质量浓度与所加入的肿瘤间质液质量浓度存在依赖性。连续培养4d间质液组对S180肿瘤细胞可见明显增殖作用,高质量浓度组最明显。结论肿瘤间质液(痰浊)对体外培养S180肿瘤有生长和增殖有促进作用,且成质量浓度依赖性。
Objective：To observe the proliferation of the S180 cancer cells with the interstitial fluid（turbid phlegm） in vitro.Methods：The tumor-bearing mice were inoculated with 0.2mL suspension which was 3 passage S180 sarcoma solid tumor with 2：1 dilution（Saline：Tumor weight）.After 2 weeks,the cancer cells interstitial fluid without tumor cells were made from the tumor and used in the treatment group with 3 concentrations.Meanwhile,the control group was used 0.9% saline.The proliferation of S180 cancer cell was observed by MTT method.Results：There was significant differences of OD value between several concentrations interstitial fluid.It was decreasing concentration relationship of these concentrations.The level of the S180 cancer cell was depended by the concentration of the interstitial fluid.There was obvious growth of the S180 cancer cell in the 4 days interstitial fluid group.The more significant proliferation,the more high concentration.Conclusion：It is concentration dependence that the proliferation of the S180 cancer cell is promoted by the tumor interstitial fluid（turbid phlegm）.
