摘要
目的:探讨黄精多糖(polygona-polysaccharose,PP)对APP转基因小鼠海马CA1区突触结构的影响及其可能机制。方法:26只APP转基因小鼠,随机分为空白对照组(n=8)、低剂量黄精多糖治疗组(n=9)和高剂量黄精多糖治疗组(n=9)。采用不同浓度黄精多糖溶液对治疗组APP转基因小鼠连续ig45d后,取海马CA1区组织在透视电镜下观察,对其突触的数量、面积、变性程度及其含有突触小泡数量进行体视学分析。结果:黄精多糖治疗组突触数量增多(P<0.01);突触截面积减小且变性程度减轻(P<0.01);突触中突触小泡数量增多(P<0.01);改善程度具有一定量效关系。结论:黄精多糖能保护APP转基因小鼠海马CA1区突触结构。
Objective:To investigate the effects of polygona-polysaccharose ( PP) upon the synapse structure in hippocampus s region of APP transgenic mice and the related mechanism. Method:Different concentrations of PP solution were Ig given to APP transgenic mice daily for 45 consecutive days,then tissues of hippocampuss CAI region were observed through transmission electron microscope ( TEM),and stereology was adopted to analyze the quantity,sectional area and degeneration of its synapse and the alteration of the quantity of the synapse vesicles involved. Result:The PP treated groups exhibited an obvious increase in synapse quantity (P 〈0. 01),a decrease in synapse sectional area and degeneration degree,and an increase in synapse vesicles quantity,and the improvement of its synapse had dose-effect relation to some extent. Conclusion:PP is beneficial for the improvement of the synapse structure in hippocampuss CAI region of APP transgenic mice.
