摘要
目的:观察头痛宁对偏头痛大鼠模型硬脑膜动脉血流和外周血中降钙素基因相关肽(calci-tontin gene-related peptide,CGRP)的调节作用。方法:24只雄性SD大鼠随机分成四组:生理盐水组(N组)、头痛宁组(T组)、舒马曲坦组(S组)、假手术组(Sham组)。电刺激三叉神经节建立偏头痛大鼠模型,放射免疫分析法测定大鼠外周血中CGRP含量,利用激光多普勒脑血流仪检测大鼠硬脑膜动脉血流量。结果:电刺激三叉神经节时,T组与N组比较其CGRP含量明显低于N组,差异非常显著(P<0.01),而与S组比较差别不大(P>0.05),N组与Sham组比较差异非常显著(P<0.01)。电刺激三叉神经节时,T组与N组比较其脑膜血流量明显低于N组,差异非常显著(P<0.01),而与S组比较差别不大(P>0.05),N组与Sham组比较差异非常显著(P<0.01)。结论电刺激三叉神经节可导致大鼠外周血中CGRP含量及脑膜血流量增加。头痛宁能明显抑制CGRP的表达与减少脑膜血流量,从而有效地治疗偏头痛。
Objective: To investigate the role of Toutongning capsule on the dura artery blood flow and CGRP level in peripheral blood of the rat models with migraine. Methods : The 24 male SD rats were randomly divided into 4 groups : normal saline treatment group ( group N), Toutongning treatment group (group T), sumatriptan treatment group (group S), sham-operated group (group sham). The rat model with migraine was manufactured by electrical stimulation of the trigeminal ganglion. The radioimmunoassay method was used to mensurate of CGRP level in rat peripheral blood. The laser Doppler instrument was applied to test rat dura artery blood flow. Results: When the trigeminal ganglion was electrically stimulated, CGRP level in peripheral blood in rats of the group T was significantly lower than that of group N (P 〈 0.01 ), which did not have any significant difference when compared with the group S (P 〉 0.05 ). And this was significantly different between group N and group sham ( after sham-operated) ( P 〈 0. 01 ). When the trigeminal ganglion was electrically stimulated, the dura artery blood flow in rats of the group T was significantly lower than that of group N (P 〈0.01 ), which did not have any significant difference when compared with the group S ( P 〉 0.05 ). And this was significantly different between group N and group sham ( after sham-operated) (P 〈 0. 01 ). Conclusion : The CGRP levels in peripheral blood and the dura blood flow increased during electrical stimulating of the trigeminal ganglion. Toutongning capsule may significantly inhibit peripheral blood CGRP, and also restraint the rat dura blood flow, so it may treat migraine effectively.
