摘要
目的制备尤利沙星光学异构体,并确定其绝对构型。方法以二甲基亚砜为溶剂,L-或D-酒石酸为拆分剂,对消旋的尤利沙星进行拆分,并测定其圆二色性光谱图。结果从消旋的尤利沙星拆分得到的光学异构体的光学纯度>95%;与1H,4H-[1,3]硫氮杂环丁烷并[3,2-a]喹啉-3-羧酸类似物的圆二色性光谱图比较,可确定尤利沙星的左旋体C-1的绝对构型是S型,尤利沙星的右旋体C-1的绝对构型是R型。结论化学法拆分制备尤利沙星光学异构体工艺具有实际应用价值,可在工业上应用。
Objective To prepare optically active isomers of ulifloxacin and establish their absolute configurations. Methods Ulifloxacin was resolved with L(+) and D(-)tartaric acid and DMSO.The absolute configurations of its optical isomers were determined by the circular dichroism spectra.Results Racemic ulifloxacin has been resolved into two optical isomers(95% ee).Their absolute configurations were determined by comparing with the circular dichroism spectra of the derivatives of 1H,4H-Thiazetoquinoline-3-carboxylic acids:(-)-ulifloxacin was S-isomer,(+)-ulifloxacin was R-isomer.Conclusion The process of chemical resolution of ulifloxacin is practically valuable and promising in industry.
