摘要 目的:研究PI3K/Akt信号转导通路介导的细胞凋亡相关因子在胃肠道间质瘤(GIST)中的表达及意义。方法:RT-PCR检测PI3K、NF-κB、XIAP、bcl-2与survivin在124例GIST及癌旁正常组织中mRNA水平。结果:在GIST中,PI3K、NF-κBmRNA表达与NIH分级、肿瘤侵袭转移、黏膜受侵密切相关,而与年龄、性别、组织学类型无关。XIAPmRNA随NIH分级表达上调;XIAP表达与GIST组织分化程度呈负相关,即分化程度越低,XIAP阳性表达率愈高,伴有肿瘤侵袭转移的GIST病例XIAP表达水平较高,同时XIAP的表达与黏膜受侵密切相关,而与年龄、性别、组织学类型无关。bcl-2mRNA水平表达上调,与NIH分级、黏膜受侵有关,而与年龄、性别、组织学类型、转移复发及周围组织浸润无关。survivinmRNA在GIST中的表达与NIH分级、肿瘤侵袭转移、黏膜受侵等因素有关,而与年龄、性别以及组织学类型无关。由于NF-κB的调控作用,各指标之间mRNA的水平变化呈正相关。结论:GIST中PI3K/Akt信号转导通路的活化可以进一步激活NF-κBmRNA,从而进入细胞核内调控凋亡相关因子XIAP、bcl-2以及survivinmRNA水平的表达。 Objective: To investigate the detection and significance of apoptosis related factors controlled by PI3K/Akt signal transduction pathway in gastrointestinal stromal tumors(GIST). Methods: The expression of PI3K,NF-κB,XIAP,bcl-2 and survivin was detected with RT-PCR using specimens from 124 cases of GIST and the corresponding normal gastric mucosa. Results: In gastrointestinal stromal tumors,the activation of PI3K/Akt signal transduction pathway can further active NF-κB mRNA,and then it can enter nucleus and control the mRNA expression of apoptosis-related factors:XIAP,bcl-2 and survivin. Conclusion: The mRNA expression of NF-κB,XIAP,bcl-2 and survivin is closely related to NIH classification,mucosal invasion and lymphatic metastasis.They have no relationship with age,sex and histological type.Based on the regulation of NF-κB regulation,the expression of them are of positive correlation.
