槲寄生碱联合顺铂对人肺腺癌A549细胞的生长抑制及诱导凋亡作用 被引量:2

摘要 目的探讨槲寄生碱、顺铂及两者联用对人肺腺癌细胞A549增殖、凋亡和细胞周期的影响。方法 MTT法观察槲寄生碱、顺铂及两者联用对人肺腺癌A549细胞增殖的影响;DAPI染色观察槲寄生碱、顺铂及两者联合作用时A549细胞凋亡形态,流式细胞术检测槲寄生碱、顺铂和两者联合应用对人肺腺癌A549凋亡和细胞周期的影响。结果①槲寄生碱、顺铂均能抑制人肺腺癌细胞A549的生长,并表现为浓度和时间的依赖性;槲寄生碱0.0625、0.125mg/mL分别与顺铂1、2mg/L联合24、48、72h的抑制率显著高于单用顺铂、槲寄生碱组(均为P<0.01),两者呈现出相加的抗瘤效果。②DAPI染色后槲寄生碱组、顺铂组和联合用药组A549肺癌细胞均表现出典型的细胞凋亡特征。③槲寄生碱和顺铂均可在一定程度上诱导细胞凋亡,两种药物联用后,诱导细胞凋亡作用显著增强(P<0.01)。④槲寄生碱将细胞周期阻滞在G0/G1期,顺铂将细胞周期阻滞在S期。结论槲寄生碱、顺铂均可抑制人肺腺癌A549细胞的增殖、诱导细胞的凋亡,槲寄生碱可明显提高顺铂对A549细胞的杀伤效果,两者具有明显的相加作用。 [Objective] To study the effect of mistletoe alkili, cisplatin and their combination on the growth, apoptosis and cell cycle distribution of human lung adenocareinoma cell hne A549. [ Methods ] The inhibitory effects of mistletoe alkili and cisplatin on the growth of A549 cells was measured by MTT assay, and apoptosis of A549 cells were observed by DAPI staining. The effects of mistletoe alkil, cisplatin and their combination on A549 cells were detected by flow cytometry to investigate the apoptosis and cell cycle distribution of lung adenoearcinoma cells. [Results] (1)Both mistletoe alkali and cisplatin could inhibit the A549 cell proliferation in a concentrationdependent and time-dependent pattern. Compared with either mistletoe alkili or eisplatin alone, 0.0625, 0.125mg/mL of mistletoe alkili combined with 1 or 2 mg/L of cisplatin respectively could increase the growth inhibition rate of A549 ceils significantly(all P 〈0.01), and the results showed an additive pattern of effects. (2)The A549 cells showed a typical features of apoptosis by DAPI staining, including mistletoe alkali group, cisplatin group and the combined treatment group. (3)Both mistletoe alkili and cisplatin could induce apoptosis of A549 cells to some extent. Especially when cisplatin was combined with mistletoe alkali, the ratio was increased significantly (P 〈0.01). (4)A549 cells showed G0/G1 arrest after mistletoe alkili administration (P 〈0.01), and S stage arrest with cisplatin administration (P 〈0.01). [ Conclusion ] Both mistletoe alkili and cisplatin could inhibit the growth and enhance the apoptosis of human lung adenocarcinoma A549 cells in a concentration-dependent pattern. When they were combined, the effect could be obviously enhanced in an additive pattern.
