摘要
目的:观察乳腺癌新辅助化疗后不同时间肿瘤细胞Ki-67表达水平的变化情况,研究乳腺癌新辅助化疗后肿瘤细胞增殖变化的规律,探讨依此指导选择化疗最佳间期的可行性。方法:将乳腺癌组织接种于裸鼠前腿根部皮下,待其成活并生长到0.3~0.4 cm时,随机分为8组,取其中1组肿瘤组织应用免疫组化法测定肿瘤细胞Ki-67表达水平,其余7组行CAF(环磷腺胺、多柔比星和5-氟尿嘧啶)方案化疗1次,分别在化疗后1、3、5、7、14、21和28 d取肿瘤组织,应用免疫组化法测定肿瘤细胞Ki-67表达水平。结果:乳腺肿瘤细胞Ki-67表达水平从化疗后第3天开始逐渐降低,至化疗后14 d时降到最低点,然后又逐渐升高,至化疗后28 d时已接近化疗后第5天水平。结论:乳腺癌新辅助化疗后肿瘤细胞的增殖有其特定的变化规律,应用免疫组化法测定化疗后肿瘤细胞Ki-67表达水平,有可能指导临床选择最佳化疗间期,从而提高乳腺癌新辅助化疗效果。
OBJECTIVE: To observe the change of the level of the Ki-67 gene in different time of breast carcinomas after preoperative chemotherapy,investigate the disciplinarian of proliferation and analyze the feasibility of chemotherapy time.METHODS: The breast fat pads of mude mice were inoculated with the survival breast tissue.When the pads grew to 0.3-0.4 cm,they were randomly divided into eight groups.Seven groups of that had chemotherapy of CAF scheme(cyclophosphamide,arubicin,and 5-fluorouracil).The expression of Ki-67 of tumor cells was detected by immunohistochemistry in the rest of one group,and in the chemotherapy-groups 1,3,5,7,14,21 and 28 days after the chemotherapy.RESULTS: The Ki-67 expression levels of breast cancer cells gradually lowered from the first three days after the chemotherapy,to bottom after 14 days,then gradually increased after 28 days,reaching nearly the level of 5 days after the chemotherapy.CONCLUSIONS: Breast cancer cell proliferation after neoadjuvant chemotherapy has its particular variation.Application of the immunohistochemical determination method for detecting Ki-67 expression levels of tumor cells after chemotherapy,may guide the best chemotherapy,thereby improving the interphase effect of breast cancer neoadjuvant chemotherapy.
