摘要
目的探讨导流杂交基因芯片技术作为临床检测人乳头瘤病毒(HPV)常规方法的实用性,及在宫颈癌筛查中的临床应用价值。方法采用导流杂交基因芯片技术对676例疑似HPV感染的标本进行基因诊断和分型,检测了13种高危亚型(16,18,31,33,35,39,45,51,52,56,58,59和68型)、5种低危亚型(6,11,42,43和44型)和3种中国人群常见亚型(53,66和CP8304型)。结果 676例样本中检出HPV阳性175例,检出率25.9%,高危占74.9%、低危占16.0%、中国人常见亚型占9.0%、2种或2种以上亚型混合感染占15.4%。结论导流杂交基因芯片技术适合于临床筛查HPV感染,能更好的检出HPV感染的基因型,为临床提供较有价值的实验资料,对宫颈癌的筛查有较大的帮助。
[Objective]To explore the practicality of flow-through hybridization and gene chip technology as the conventional method for clinical detection of human papilloma virus (HPV), and the clinical value in the cervical carcinoma screening.[Methods] Gene diagnosis and typing were performed in 676 samples of suspected HPV infection by flow-through hybridization and gene chip technology. 13 kinds of high-risk subtypes (model l6,18,31,33,35,39,45,51,52,56,58,59 and 68), 5 kinds of low-risk subtypes (model 6,11,42,43 and 44) and 3 kinds of common subtype in Chinese population (model 53,66 and CP8304) were detected.[Results] 175 cases of HPV positive were detected in 676 samples, the detection rate was 25.9%, in which high-risk subtypes accounted for 74.9% and low-risk subtypes accounted for 16.0%. The common subtype in Chinese population accounted for 9.0%,and mixed infection caused by 2 or more kinds of subtypes accounted for 15.4%.[Conclusion]The flow-through hybridization and gene chip technology is suitable for clinical screening of HPVinfection, can detect the genotype of HPVinfection efficiently, provide the valuable experimental data for clinic, and may be helpful to screening for cervical carcinoma.
