摘要 目的:探讨非霍奇金淋巴瘤(NHL)石蜡组织切片中环氧合酶2(COX-2)和缺氧诱导因子1α(HIF-1α)的表达及两者的相关性和临床意义。方法:应用SP免疫组化法检测42例NHL和20例反应性增生淋巴结石蜡组织切片中COX-2和HIF-1α的蛋白表达情况。结果:NHL中COX-2高表达(52.38%),明显高于反应性增生淋巴结炎(10.00%),P<0.05;COX-2的表达与NHL的Ann Arbor分期显著相关,P<0.05;HIF-1α在NHL中的表达率(66.67%),明显高于反应性增生淋巴结炎(15%),P<0.05;HIF-1α的表达与NHL的Ann Arbor分期显著相关,P<0.05;NHL中COX-2与HIF-1α的表达呈中等强度正相关性(P<0.05,rs=0.539)。结论:COX-2和HIF-1α的过度表达在非霍奇金淋巴瘤的发生、发展、侵袭、转移等生物学行为中具有重要的作用,可作为临床监测非霍奇金淋巴瘤进展的指标之一,可能成为肿瘤分子靶向治疗中新的靶点。 Objective:To investigate the expression,relationship and clinical significance of cyclooxygenase-2(COX-2) and hypoxia-inducible factor-1 alpha(HIF-1α) in non Hodgkin lymphoma(NHL). Methods:Immunohistochemistry of streptavidin-peroxidase (SP) method was used to detect the expression of COX-2 and HIF-1α in 42 cases of NHL and paraffin-embodied samples of 20 cases with reactive hyperplasia of the lymph node.Results:Both of COX-2 and HIF-1α were positively expressed in NHL as compared with the reactive hyperplasic lymphadenitis tissues(52.38% vs. 10.00%,66.67% vs. 15%,respectively,P0.05) and associated with Ann Arbor staging. COX-2 showed moderate correlation with HIF-1α in NHL (P0.05,rs=0.539). Conclusion:Over-expression of COX-2 and HIF-1α may be involved in tumorigenesis,progression,invasion and metastasis of NHL and the two factors may be served as biomarkers for estimating the development of this disease and new molecular target in cancer therapy.
