Determination of Bisphenol A in Mice Tissue by High Performance Liquid Chromatogram with Florescence Detector

摘要 建立了高效液相色谱-荧光检测器测定小鼠组织中的双酚A的分析方法。小鼠组织样品经甲醇-乙酸铵缓冲液匀浆、正己烷-乙醚混合溶剂提取、40℃水浴蒸干后用流动相溶解,以乙腈-0.01mol/L乙酸铵缓冲液(pH=4.5)(体积比为75∶25)为流动相,经C18色谱柱分离,在激发波长230nm、发射波长315nm下进行荧光检测。该方法线性范围是0—20mg/L,精密度是1.46%—6.69%,回收率是80.0%—88.2%,可以满足化学品的分析要求。 A method for determination of bisphenol A in mice tissue was established.The mice tissue samples were homogenized by a high-speed homogenizer with methanol-ammonium acetate buffer.The operating conditions was as follows:C18 column(250mm×4.6mm i.d,5μm),acetonitrile-ammonium acetate buffer (pH=4.5)(75:25,V/V) as mobile phase,fluorescence detector with the excitation and emission wave length at 230nm and 315nm,respectively.The linear range of this method is from 0 to 20mg/L,the precision is from 1.46% to 6.69%,recovery is from 80.0% to 88.2%,respectively.This method could meet the need for analysis chemistry agent in environment.
