黄连对大鼠肝脏全基因表达谱的影响 被引量:5

摘要 目的:利用基因芯片技术研究寒性中药黄连对正常大鼠肝脏全基因表达谱的影响,从基因水平分析黄连作用机制。方法:SPF级Wistar雄性大鼠20只,随机分为黄连组和空白对照组,黄连组灌胃黄连水煎液10ml/kg,对照组予等容量生理盐水。给药20d后,摘取肝脏,应用RatRef-12基因芯片检测各组大鼠肝脏基因表达,筛选差异表达基因,进行基因功能归类。结果:黄连组与正常组比较有399条差异表达基因,可进行基因注释者237条,其中49.37%基因参与代谢,10.54%基因与防御反应相关,10.13%基因与免疫反应相关,10.13%基因参与氧化还原反应,6.75%基因参与炎症反应,9.70%基因参与细胞凋亡,3.38%基因参与白细胞趋化作用。结论:寒性中药黄连可上调炎性反应、免疫反应、防御反应相关基因,增强抗炎、防御能力,调控代谢、凋亡相关基因,影响P450家族成员,发挥代谢抑制、抗炎作用。 Objective:To study the effects of Coptis chinensis on whole expression profiles of liver in rats by using the gene chip technology,and analyze the mechanism of Coptis chinensis on gene level.Methods:Twenty male Wistar rats of SPF level were randomly divided into Coptis chinensis group and control group.Coptis chinensis group was given 10 ml/kg Coptis chinensis decoction and the control group given the same volume saline.After 20 days,the livers of each group were taken out to detect the gene expression by using Illumina's RatRef-12 gene array.Results:There were 399 strips of gene differential expression between the two groups,237 of which could be annotated.Among the 237 genes,49.37% were related to metabolism,10.54% to defence reaction,10.13% to immune response,10.13% to redox reactions,6.75% to inflammation,9.70% gene to apoptosis and 3.38% to leukocyte chemotaxis.Conclusion:Coptis chinensis can increase the expression of genes related to inflammatory response,immune response and defense response,enhance anti-inflammatory and defense capabilities,regulate the metabolism and apoptosis-related genes,affect the P450 family members to inhibit the metabolism and avoid inflammatory response.
