摘要
目的:研究重组人肿瘤坏死因子α(TNF-α)对放烧复合伤小鼠骨髓有核细胞(BMNC)IL-3R的调节作用。方法:以放烧复合伤小鼠为模型,观察一次性静脉注入不同剂量的TNF-α对BMNC增殖的影响及10万UTNF-α对IL-3Rα表达及功能、BMNC数量的调节作用。结果:①静注不同剂量的TNF-α对放烧复合伤小鼠BMNC增殖的影响不一;②10万U的TNF-α1、3d上调放烧复合伤小鼠IL-3Rα的平均含量(对阳性率及功能无明显影响),而致BMNC数量明显降低;7d时恢复至对照组水平;③10万UTNF-α1、3d致放烧复合伤小鼠BMNC计数明显降低。结论:10万UTNF-α在体内虽能上调放烧复合伤小鼠IL-3Rα的平均含量而剌激造血,但可能因其同时激活造血调控网络整体效应却抑制骨髓造血。
Objective: To investigate the effects of recombinant human tumor necrosis factor α
(RHTNF α) on interleukin 3 receptor (IL 3R) of bone marrow nucleated cells (BMNCs) after
combined radiation burn injury in mice. Methods: After the combined radiation burn injury was
inflicted on the mice, the effects of RHTNF α at different doses on proliferation of BMNCs and
the regulative effects of a single injection of 10 5U of RHTNF α on the expression and
function of IL 3R α and BMNC counts were determined. Results: ①Different doses of RHTNF
α exerted different effects on the proliferation of BMNCs in vivo. ②The mean content of IL
3R α was up regulated on the 1st and 3rd d but recovered to the normal on the 7th d after the
injection of 10 5 U of RHTNF α. ③The single injection of 10 5 U of RHTNF α caused a rapid
decrease in BMNC counts on the 1st day after injection and the counts remained low up to the
3rd. Conclusion: A single injection of 10 5 U of RHTNF α can up regulate the expression of IL
3R of BMNCs after combined radiation burn injury, which stimulates hematopoiesis. However,
it causes a significant decrease in BMNC counts because of the effects of the hematopoietic
network.
