白藜芦醇对模拟日光照射后黑素和朗格汉斯细胞的影响 被引量:2

摘要 目的:观察白藜芦醇对模拟日光照射后黑素和朗格汉斯细胞的影响。材料和方法:招募11名健康女性志愿者,在每位志愿者背部选取6个非曝光部位。部位1~4分别涂抹白藜芦醇+抗氧化剂、抗氧化剂、白藜芦醇、基质,30min后予1.5倍最小红斑量的模拟日光照射,部位5仅予照射而不涂抹检测物质(阳性对照),部位6既不照射亦不涂抹检测物质(阴性对照)。每天照射一次,连续照射4天。末次照射后24h对皮肤进行取材,进行Fontana Manna染色及CD1a免疫组化染色。结果:阳性对照部位和涂抹基质部位黑素含量指数(melanin content index,MCI)-目标面积/场面积比值(%)及平均光密度值显著上升,朗格汉斯细胞显著减少;而双侧涂抹白藜芦醇+抗氧化剂、抗氧化剂、白藜芦醇的部位MCI-目标面积/场面积比值(%)及平均光密度值轻度上升,朗格汉斯细胞轻度减少,与阳性对照部位和涂抹基质的部位相比具有显著差异(P<0.05)。结论:联合应用白藜芦醇和抗氧化剂或单独应用二者之一均可有效地防护模拟日光照射引起的黑素颗粒增加和朗格汉斯细胞数量减少。 Objective To investigate the effects of resveratrol on melanin and Langerhans cells from solar simulator ultraviolet irradiation (ssUVR).Materials and Methods Eleven healthy female volunteers were enrolled in this study.Six sites on the non-exposed back skin were marked for the experiment.Sites 1~4 were applied with resveratrol (Res)+antioxidant (Aox),antioxidant,resveratrol,vehicle,and followed by ssUVR at a dosage of 1.5 minimal erythema dose (MED) 30 min later.Site 5 received ssUVR without any test material (positive control)and site 6 received neither ssUVR nor test materials (negative control).The skin biopsies were taken 24 hours after the last ssUVR.The specimens were stained with Fontana Manna staining and immunohistochemistry staining of Monoclonal Antibody-CD1a.Results The measured percentage (%) and average optical density of MCI increased significantly on UV only site and Vehicle+UV site,much higher than on Res+Aox+UV site,Aox+UV site and Res+UV site (P〈0.05).The density of Langerhans cells decreased dramatically on UV only and Vehicle+UV site,significant more than on Res+Aox+UV site,Aox+UV site and Res+UV site (P〈0.05).Conclusions Resveratrol,antioxidant or their combination were effective in protecting the skin from ssUVR-induced melanin increase and Langerhans cells depletion.
