The Influence of 2-Ethylhexyl Phthalate on Testicular Synthesis and GATA-4 mRNA Expression in Mice

摘要 目的研究邻苯二甲酸二乙基己酯对大鼠睾丸睾酮合成、GATA-4 mRNA表达的影响。方法雄性SD大鼠32只,随机分成4组,每组8只,设3个染毒剂量组(10,100,500mg/kg.d DEHP溶媒为玉米油)和1个对照组(玉米油),饲养1个月后处死。酶联免疫吸附法测定血清中睾酮含量;RT-PCR法检测睾丸组织GATA-4 mRNA表达的含量。结果实验组血清中睾酮含量和睾丸组织的GATA-4 mRNA含量均低于对照组。结论邻苯二甲酸二乙基己酯可使大鼠睾丸Leyd ig细胞变性,血清睾酮合成降低,其机制可能与降低GATA-4 mRNA表达有关。 Objective To study the change of testosterone synthesis function of interstitial cell ( Lcydig cell) and the expression GATA -4 mRNA in mice exposed to di (2 - ethylhexyl) phthalate (DEHP). Methods 32 male Sprague - Dauley rats were divided into four groups randomly: normal control group and 3 groups exposed to DEHP at the dose of 100, 200 or 500 mg/kg bodyweight per day by garage respectively. One month later, the rats were sacrificed. The serum testosterone (T) concentration was measured by ELISA. The mRNA expression level of GATA - 4 mRNA of testicular tissue were detected by RT - PCR. Results The mRNA levels of GATA - 4 of testicular tissue and the serum testosterone concentrations decreased in 3 groups compared with normal control group. Conclusion DEHP can downregulate the serum testosterone concentration. The decreasing mRNA expression of GATA -4 may be involved in it.
