摘要
目的观察红色毛癣菌在盐酸小檗碱作用下形态学变化。方法应用美国CLSI制订的标准M38-A方案,采用MTT微量稀释法测定盐酸小檗碱对红色毛癣菌的体外抑制率。将盐酸小檗碱作用前后的红色毛癣菌制成标本,分别在扫描电子显微镜和透射电子显微镜下观察形态学变化。结果盐酸小檗碱作用于红色毛癣菌后,扫描电子显微镜下菌丝表面变粗糙,菌丝萎缩、皱瘪、粗细不一,出现多处破损、断裂和小的破坏性孔洞。透射电子显微镜下细胞变形,呈不规则形;细胞壁粗糙,厚薄不均;细胞浆结构模糊,细胞核凝固、破碎。结论盐酸小檗碱使红色毛癣菌的形态发生明显变化。
Objective To observe morphological changes of Trichophytosis rubrum treated by berberine hydrochloride.Methods Application MTT micro-dilution method, standard M38-A of the United States CLSI was used to evaluate the inhibition of berberine hydrochloride on Trichophyton rubrum in vitro. Before and after Trichophyton rubrum specimens was treated by berberine hydrochloride respectively, the morphological changes were observed by scanning electron microscopy and transmission electron microscope. Results After berberine treated, Trichophyton rubrum＇s hyphae surface showed rough, shrunken, wrinkle deflated and thickness varies accompany with broken, fractured and small destructive holes under scanning electron micro- scope. The photo of transmission electron microscope showed that the hyphae became deformed, which presented the irregular form, their cell wall was roughly and loosely, cytoplasm texture was obscured and cellular nucleus was syneresis and quassation. Conclusion Morphology of Trichophytosis rubrum treated by berberine hydrochloride change obviously.
