子痫前期患者外周血HLA-G水平及CD4^+CD25^+Foxp3^+/CD4^+T细胞亚群比例与眼底病变的关系研究

摘要 探讨子痫前期患者外周血HLA-G水平及调节性T细胞亚群的变化与眼底病变的关系。分别采用ELISA法和流式细胞技术检测28例住院子痫前期患者(轻、重组)产前和产后血清HLA-G水平和外周血T细胞CD4^+CD25^+Foxp3^+/CD4^+比值,并引入14例正常妊娠待产产妇作对照组,对比分析各组HLA-G水平及CD4^+CD25^+Foxp3^+/CD4^+与眼底病变的关系。结果不同病情子痫前期患者之间产前HLA-G水平具有统计学意义(P<0.01),产前、产后不同病情子痫前期患者CD4^+CD25^+Foxp3^+/CD4^+具有统计学意义(P<0.01),并且随着病情加重而降低;不同程度眼底病变之间产前HLA-G水平具有统计学意义(P<0.01)产前、产后不同程度眼底病变之间CD4^+CD25^+Foxp3^+/CD4^+具有统计学意义(P<0.01),并且随眼底病变加重呈下降趋势。产前、产后眼底病变与病情程度之间有显著相关性(P<0.01)。子痫前期患者外周血HLA-G水平及CD4^+CD25^+Foxp3^+/CD4^+是反映子痫前期病情和眼底视网膜病变程度的指标之一。 To detect the levels of HLA-G and CD4+CD25+Foxp3+/CD4+ T cells in peripheral blood and their relation to retinopathy in preeclampsia patients,enzyme-linked immunosorbent assay(ELISA) and flow cytometry were adopted to detect the concentration of HLA-G in plasma and the rate of CD4+CD25+Foxp3+/CD4+ T cells in peripheral blood of 28 cases of women with pre-eclampsia classified into two groups by the extent of disease before and after delivery.In addition,and 14 cases of women with normal pregnancy were included serving as control group.The indexes were compared,and the correlations with retinopathy were discussed.It was found that there was apparent statistical difference in the level of HLA-G among pre-eclampsia patients before delivery(P0.01).The concentration decreased with the state of illness getting worse.And the apparent statistical consequences before and after delivery were acquired when it came to be the CD4+CD25+Foxp3+/CD4+ T cells(P0.01).Also,the level of HLAG in plasma showed statistical difference among three groups classified by the extent of retinopathy before delivery(P0.01),and the index declined when the stage of retinopathy developed.The same situation occurred before and after delivery during referring to CD4+CD25+Foxp3+/CD4+ T cells,indicating there were conspicuous relationship between the extent of retinopathy and the disease(P0.01).In conclusion,it is apparent that the lever of HLA-G and the rate of CD4+CD25+Foxp3+/CD4+ T cells in peripheral blood may play an important role in pathogenesis of retinopathy and are the good index to reflect the extent of retinopathy in preeclampsia.
