摘要 目的:研究重度子痫前期患者外周血中性粒细胞的凋亡及其机制。方法:分离培养20例重度子痫前期患者外周血中性粒细胞,并设立正常妊娠待产产妇20例为对照组。培养24h后通过形态学观察和流式细胞术测定凋亡率分析中性粒细胞的凋亡;用免疫细胞化学方法检测中性粒细胞Bax蛋白的表达。结果:两组中性粒细胞均呈现凋亡极具特征性的形态学改变;重度子痫前期组中性粒细胞凋亡率48.57%明显低于正常妊娠待产组66.67%,差异有统计学意义(P<0.01);重度子痫前期组外周血中性粒细胞免疫化学染色显示Bax蛋白表达量与凋亡率呈强正相关(r=0.886,P<0.01)。结论:重度子痫前期患者外周血中性粒细胞存在凋亡延迟,其机制可能与Bax蛋白表达减少有关。 Objective:To investigate the peripheral neutrophil apoptosis in severe preeclampsia and its mechanism. Method: Neutrophils isolated from 20 severe preeclampsia patients and 20 normal pregnancies were incubated in vitro for 24h. Then apoptosis was assessed by morphologic changes ,and neutrophils apoptosis rate was determined by flow cytometry. Production of proapoptotic protein in Bax by neutrophils was evaluated by immunocytochemistry. Result:The neutrophils of the two groups exhibited morphologic evidence of apoptosis. The apoptotic rate of severe preeclampsia was significantly lower than that of control group (48.57 % vs 66.67% ,P 〈0.01 ). In all groups,the Bax immuno reactivity was strongly related with apoptotic neutrophils( r = 0. 886 ,P 〈 0.01 ). Conclusion: Apoptosis in peripheral neutrophil of severe preeclampsia delays. The reduced expression of Bax may be one of the possible inner mechanisms.
