子宫腺肌病在位、异位内膜中EST和COMT的表达及其相关性 被引量:1

Expression of Estrogen Sulfotransferase and Catechol-O-methyl Transferase in Eutopic and Ectopic Endometria in Adenomyosis and Their Correlation

摘要 目的比较雌激素硫酸转移酶(estrogen sulfotransferase,EST)、儿茶酚氧位甲基转移酶(catechol-O-methyl transferase,COMT)在子宫腺肌病异位内膜、在位内膜与正常子宫内膜中表达的差异,探讨其在子宫腺肌病发生发展过程中的意义。方法采用免疫组化链霉素-生物素-过氧化物酶复合物法(SABC法)检测40例子宫腺肌病患者的异位内膜及相应的在位内膜以及40例对照组正常内膜中EST、COMT的表达水平,探讨其是否有差异性表达,并分析同一内膜不同因子表达的相关性。结果①EST在子宫腺肌病异位内膜、在位内膜及对照组正常子宫内膜中的表达差异均无统计学意义(均P>0.05);②COMT在子宫腺肌病异位内膜表达的平均吸光度值高于在位内膜及对照组正常子宫内膜,差异有统计学意义(均P<0.05);③EST、COMT在异位内膜及在位内膜中的表达无相关性,两因子在正常子宫内膜增生期及分泌期的表达均呈负相关性。结论 COMT在子宫腺肌病异位内膜的雌激素代谢中有着重要的作用,并且可能与局部雌激素水平的调节有关,对于子宫腺肌病的发生及发展有着重要的作用。 Objective To investigate the expression of estrogen sulfotransferase(EST) and catechol-O-methyl transferase(COMT)in adenomyosis and their correlation.Methods The expression of EST and COMT was detected by immunohistochemistry(SABC method)in ectopic and eutopic endometria of 40 cases of adenomyosis,and normal endometria of 40 cases of patients with cervical intraepithelial neoplasia grade 3(CINⅢ).Results The average absorbance values of the EST in ectopic and eutopic endometria of patients with adenomyosis were insignificantly lower than those in the normal endometria of patients with CINⅢ(all P〉0.05);The average absorbance values of COMT expression in ectopic and eutopic endometria of patients with adenomyosis were significantly higher than those in the normal endometria of patients with CINⅢ(P〈0.05);There was no correlation between the expression of EST and COMT in the ectopic endometria,but in normal endometria there was a negative correlation between them.Conclusion The EST levels had no significant change.The over-expression of COMT in the ectopic endometria of the patients with adenomyosis has a close relation with the metabolism pathogenesis of estrogen in the ectopic endometria,which may play a key role in the pathogenesis and development of adenomyosis.
