摘要
目的研究电针督脉腧穴对脑缺血大鼠缺血灶周围巢蛋白表达的影响。方法 48只Wistar大鼠随机分为正常组、模型组和电针组,每个组又细分为7d组和14d组2个亚组。制备大脑中动脉栓塞模型,电针百会和大椎穴。免疫组化染色方法和蛋白印迹技术定量分析不同时间点、不同组别之间缺血灶周围巢蛋白表达的差异。结果与模型组比较,电针组巢蛋白表达增高,且电针组7d时巢蛋白反应条带的相对灰度值最高,14d时巢蛋白表达下降。结论电针督脉腧穴可以激发脑缺血大鼠缺血灶周围巢蛋白表达,促进神经元再生。
Objective To investigate the effect of electroacupuncture(EA)on acupoints of Du Meridian on the expression of Nestin around the ischemic loci in rats with cerebral ischemia.Methods Forty-eight healthy male Wistar rats were divided into normal,model and EA groups randomly.Furthermore,rats in each group were divided into 7-day and 14-day subgroups.Model of middle cerebral artery occlusion(MCAO)was established,then EA for 7 days or 14 days on acupoints of Baihui and Dazhui.The expression of Nestin around the ischemic loci was detected at different time points and in different groups by immunohistochemical technique and Western blot.Results The expression of Nestin in EA group was significantly increased as compared with model group.Much more Nestin-immunoreactive cells were observed in EA group on the day 7 and the expression was decreased on the day 14.Conclusion These findings indicate that EA on acupoints of Du Meridian has the potential to activate Nestin around the ischemic loci of rats with cerebral ischemia and facilitate neuroregeneration.
