摘要
目的:观察补肾益气方药益气聪明汤、加减益气聪明汤对D-半乳糖致衰老模型大鼠脑组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量及海马PKA、CREB、c-fos蛋白表达的影响。方法:雄性SD大鼠随机分为青年对照组、D-半乳糖致衰老组(简称衰老组)、益气聪明汤组及加减益气聪明汤组。除青年对照组大鼠外,其余各组大鼠以大剂量D-半乳糖腹腔注射制造衰老模型。Morris水迷宫法观察大鼠空间学习记忆能力;分光光度法检测大鼠脑组织SOD活性以及MDA含量;Western-blot法检测大鼠海马PKA、CREB、c-fos蛋白表达的变化;同时观察补肾益气方药对衰老模型大鼠上述指标的影响。结果:与青年对照组相比,衰老组大鼠脑SOD活力明显降低(P<0.05),MDA含量显著升高(P<0.05),PKA、CREB、c-fos的蛋白表达水平明显降低(P<0.05)。与衰老组大鼠相比,益气聪明汤组和加减益气聪明汤组可改善衰老模型大鼠退化的空间学习记忆能力(P<0.05);提高大脑SOD活力,降低MDA含量(P<0.05);显著上调衰老模型大鼠海马组织PKA、CREB、c-fos的蛋白表达水平(P<0.05)。结论:补肾益气方药益气聪明汤、加减益气聪明汤可能是通过调节与学习记忆相关的cAMP/PKA/CREB信号转导通路关键分子(PKA、CREB、c-fos)的表达,从而改善衰老大鼠空间学习记忆能力,延缓衰老。
Objective:To observe the effects of "Yiqi Congming Decoction" and "Jiajian Yiqi Congming Decoction" on SOD activity, MDA content and the expressions of PKA, CREB, c-fos in hippocampus of aging rats induced D-galactose. Methods:The SD male rats were randomly divided into young eoutrol group, model group, Yiqi Congming Decoction group and Jiajian Yiqi Congming Decoction group. The aging model of rat was established by injection of Dgalactose. Morris water maze was used for measuring the spatial memory, spectropho meteric analysis was used for determining the activity of SOD and the content of MDA in the cortex and Western-blot for detecting protein expressions of PKA, CREB and c-fos in hippocampus. The influences of reinforcingkidney and replenishing-qi recipe on above indexes of the aging model were also observed. Results: Compared with young control group, in model group the activity of SOD decreased significantly ( P 〈 0.05 ), the content of MDA increased significantly ( P 〈 0.05), and the protein expressions of PKA, CREB, and c-fos decreased significantly ( P 〈 0.05). Compared with aging rat group, in the Yiqi Congming Decoction group and Jiajian Yiqi Congming Decoction group, the rats' spatial memory were improved significantly (P 〈 0.05 ), the activity of SOD increased, the content of MDA decreased significantly (P 〈 0.05 ), and the protein expressions of PKA, CREB, and c-fos increased significantly ( P 〈 0.05 ). Conclusion : "Yiqi Congming Decoction" and "Jiajian Yiqi Congming Decoction" can improve aging rats' spatial memory ability and delay aging through regulating the expressions of PKA, CREB and c-fos, the key molecules in cAMP/PKA/CREB signal transduction pathway related to learning and memory ability.
