摘要
目的探讨兔耳软骨细胞与聚羟基丁酯-聚丙交酯复合物(PHB-PLA)的细胞相容性,为进一步以PHB-PLA为支架构建组织工程软骨提供实验依据和理论基础。方法分离、培养的兔耳软骨细胞,接种于PHB-PLA浸泡液及膜片上,四甲基偶氮唑蓝(MTT)法测定细胞增殖指数及计算毒级,并在体外培养后组织染色(Masson)及扫描电镜观察细胞的生长状况。设立空白对照组。结果兔耳软骨细胞在PHB-PLA浸泡培养基中生长良好。24、487、2 h MTT法结果与对照组差异无统计学意义;毒性分级为0。组织染色及扫描电镜显示:兔耳软骨细胞生长良好,形态正常,黏附牢固。结论兔耳软骨细胞与PHB-PLA的相容性良好,无毒性,可以用于进一步构建组织工程软骨。
Objective To study the cyto-compatibility between chondrocytes of auricular cartilage in rabbits and the blending of PHB with PLA,in order to provide experimental evidence and theoretical foundation for fabricating tissue engineering cartilage using PHB-PLA as scaffold.Methods We cultured the chondrocytes isolated from auricular cartilage of New Zealand rabbits in vitro.Proliferate and harvest the chondrocytes,and then inoculate them at soak fluid and the chaff of PHB-PLA.Proliferation index of cells was detected with MTT assay,and the growing condition of cells was examined with scanning electron microscope(SEM).Results The chondrocytes of auricular cartilage in rabbits grew well in PHB-PLA medium.No signficant difference was found between the results of 24 hours,48 hours and 72 hours MTT assay and that in the control group.The toxicity gradation of PHB-PLA was 0.The histological and electron microscopic examination revealed that chondrocytes of auricular cartilage in rabbits were in well growth and eumorphism with firm adherence.Conclusion The chondrocytes of auricular cartilage in rabbits have good compatibility with PHB-PLA,good adherence,and can be used to build further tissue engineering cartilage.
