人磷酯酰蛋白聚糖3N端融合蛋白多克隆抗体制备及鉴定 被引量:2

摘要 目的:制备人磷酯酰蛋白聚糖3(Glypican3,GPC3)N端蛋白的多克隆抗体。方法:在大肠埃希菌Rosetta中诱导表达磷酯酰蛋白聚糖3N端融合蛋白,从SDS-PAGE中切胶纯化,免疫新西兰兔,制备多克隆抗体。用辛酸-硫酸铵分步沉淀法初步纯化抗血清,并用ELISA、Westen-blot检测抗体的灵敏度和特异性。结果:成功制备了抗磷酯酰蛋白聚糖3N端的多克隆抗体,抗体滴度为1:64000,经Western blot检测特异性良好。结论:成功制备了抗人磷酯酰蛋白聚糖3N端融合蛋白多克隆抗体。 Objective:To prepare rabbit anti-human Glypican3 N-terminal protein polyclonal antibody.Methods:The Glypican3 N-terminal protein was expressed in E.coli Rosetta.The rabbits were immunized by purified protein to produce polyclonal antibody.The anti-sera were identified with ELISA and Western blot analysis.Results:The polyclonal antibody against Glypican3 N-terminal protein was successfully prepared and the titer of antisera was 1:64 000.Conclusion:The polyclonal antibody against human Glypican3 N-terminal protein was successfully prepared.
