免疫磁株法联合荧光原位杂交检测微小残留病

Detection of minimal residual disease by magnetic cell sorting and fluorescence in situ hybridization

摘要 目的探讨联合免疫磁珠分选和荧光原位杂交(FISH)技术检测慢性粒细胞白血病(CML)患者微小残留病的敏感性。方法采集10例CML患者的骨髓,利用免疫磁珠法纯化CD34阳性细胞,对比分析纯化前后FISH检测结果。结果 CD34阳性细胞纯化前占细胞百分数为(0.7±0.2)%,纯化后为(93.4±2.5)%(P<0.05)。CD34阳性细胞纯化前FISH检测4例阳性,纯化后9例阳性(P<0.05)。结论免疫磁珠技术联合FISH技术可提高检测CML患者微小残留病的敏感性。 Objective To explore the sensitivity of detecting the minimal residual disease (MRD) in patients with chronic myeloid leukemia(CML) by magnetic cell sorting and fluorescence in situ (FISH).Methods The samples of bone marrows were taken from 10 CML patients.CD34 positive cells were purified by magnetic cell sorting and detected by FISH before and after purification.The results were compared by statistical analysis. Results The purity of CD34 positive cells was (0.7±0.2)% before and (93.4±2.5)% after purification(P<0.05).With FISH examination,4 cases were positive before and 9 cases were positive after purification(P<0.05).Conclusion The sensitivity of detecting MRD may be improved by combined use of magnetic cell sorting and FISH in patients with CML.
