摘要
目的探讨急性髓细胞白血病(AML)患者c-FLIPL、c-FLIPS及DLK1基因的表达水平及临床意义。方法应用逆转录-PCR半定量检测8例AML(AML组)及3例非恶性血液病患者(对照组)骨髓单个核细胞中c-FLIPL、c-FLIPS及DLK1 mRNA的表达水平。结果 AML组患者DLK1 mRNA、c-FLIPL mRNA、c-FLIPS mRNA表达均明显高于对照组(P<0.05),且AML各FAB亚型间差异无统计学意义(P>0.05)。结论 DLK1、c-FLIPL及c-FLIPS基因在AML患者中表达异常增高,可能在白血病细胞逃脱凋亡、无限增殖中发挥作用。
Objective To investigate the expression and clinical significance of DLK1 mRNA,c-FLIPL and c-FLIPS mRNA in acute myelocytic leukemia(AML) patients. Methods The expressions of c-FLIPL,c-FLIPS and delta drosophila homolog-like 1(DLK1) mRNA in bone marrow mononuclear cells (BMNNC) of 8 patients with AML(group AML) and 3 controls(group C) were detected by RT-PCR.Results The expression of DLK1 mRNA was up-regulated in group AML as compared with that in group C(P0.05).There was no significant difference in expression of DLK 1 mRNA among the subtypes of AML(P0.05).The expression of c-FLIPL mRNA was higher in group AML than that in group C(P0.05).There was no significant difference in expression of c-FLIPL mRNA among the subtypes of AML(P0.05).Furthermore,the expression of c-FLIPS mRNA was higher in group AML as compared with that in group C(P0.05),which was not significantly different among the subtypes of AML(P0.05).Conclution The expressions of DLK1,c-FLIPL and c-FLIPS mRNA in AML patients are abnormal,which may play a critical role in escaping malignant clone of AML from apoptosis and acquiring the ability to divide unlimitedly.
