实时荧光定量PCR检测急性髓细胞白血病患者MDR1基因表达及临床意义 被引量:1

摘要 目的定量分析急性髓细胞白血病(AML)患者MDR1基因的表达水平及其与临床特征及疗效的关系。方法构建实时荧光定量PCR检测MDR1基因表达的技术,并定量检测43例AML患者MDR1基因的表达水平及分析其与血象、免疫分型及临床疗效的关系。结果建立的实时荧光定量PCR方法的标准曲线相关系数>0.99。AML患者MDR 1基因表达水平较正常对照组显著升高(P<0.001),FAB分型中M2,M5型患者MDR1基因表达水平显著高于M3型患者(P<0.05及P<0.025)。MDR 1基因的表达量与AML发病时外周血白细胞计数、血红蛋白、血小板无相关关系;而伴有CD34表达阳性AML患者MDR1基因表达水平显著高于阴性患者(P<0.01);MDR 1基因高表达的初治AML患者诱导化疗的CR率显著低于低表达病例(P<0.01)。结论实时荧光定量PCR技术检测急性白血病患者MDR 1基因表达量可更准确判断AML患者的预后及早期预测难治和复发。
