摘要
目的观察延胡索碱预处理对大鼠心肌缺血再灌注心律失常的干预作用。方法将清洁级成年SD大鼠60只随机分为6组(延胡索碱高、中、低剂量预处理组,经典缺血预处理组,注射用水组和假手术组),结扎左冠状动脉前降支近段30min后再灌注60min,用心电图机监测大鼠心电变化,记录再灌注心律失常的出现时间、持续时间以及发生率,同时记录再灌注20min及60min的ST段变化。结果延胡索碱中、高剂量组的室速和室颤的发生率显著降低,与模型组比较差异有显著性(P<0.05,P<0.01)。延胡索碱低、中、高剂量组心律失常的出现时间明显推迟、持续时间明显缩短,与模型组比较差异有显著性(P<0.01)。与模型组比较,缺血预处理组和延胡索碱预处理各剂量组ST段抬高幅度无显著性差异(P>0.05),但能明显减慢心率,差异有显著性(P<0.01)。结论延胡索碱预处理可减慢大鼠心肌缺血再灌注损伤后增快的心率,还可推迟再灌注心律失常的出现时间,缩短其持续时间,降低室速和室颤的发生率。
Objective To observe the effect of capaurine pretreatment on myocardial ischemia-reperfusion arrhythmia in rats.Methods Sixty clean adult SD rats were randomly divided into model group,sham-operation group,ischemia pretreatment group,and high-,moderate-and low-dose capaurine pretreatment groups.After ligation of ramus descendens anterior arteriae coronariae sinistrae for 30 minutes and reperfusion for 60 minutes,the changes of electrocardiogram were observed,and the onset time,duration and frequency of reperfusion-induced arrhythmia were recorded.Meanwhile,the change of ST segment was observed after reperfusion for 20 and 60 minutes.Results The incidences of ventricular tachycardia（VT）and ventricular fibrillation（VF）were significantly reduced in moderate-and high-dose capaurine pretreatment groups,the differences being significant as compared with the model group（P 0.05 or P〈0.01）.In capaurine pretreatment groups,the onset time of arrhythmia was obviously postponed and the duration was obviously shortened,the differences being significant as compared with the model group（P〈0.01）.The increase of ST segment in ischemia pretreatment group and capaurine pretreatment groups did not differ from that in the model group（P〈0.05）,but ischemia pretreatment and capaurine pretreatment slowed down the heart rate（P〈0.01 compared with the model group）.Conclusion Capaurine pretreatment can slow down the increased heart rate and postpone the onset time of arrhythmia,and decrease the incidences of VT and VF.
