摘要 以根瘤土壤杆菌LNUB335为出发菌株,以维生素K3和NaN3双抗性为筛选标记,在根瘤土壤杆菌中首次利用紫外线及微波联合诱变处理,获得1株生产性能比LNUB335显著提高的突变株ARN007,其CoQ10产量为12.01mg/L,较出发菌株提高68.67%,每克干细胞含CoQ102.46mg,较出发菌株提高38.20%。通过传代实验证明该突变株的遗传性稳定,可作为进一步研究的实验菌株。 Agarobacterium tumefaciens(At)LNUB335 was used as a starting strain,with double resistance of sodium azide and vitamin K3 as screening label,mutagenized with UV combined with microwave,it was for the first time for At to be mutagenized with UV combined with microwave;and obtained a mutant strain ARN007 with production performance marked higher than LNUB335 on production of coenzyme Q10 at 12.01 mg/L,increased by 68.67% as compared with the starting strain,and the Q10 content in dried cells reached 2.45 mg/g,increased by 38.20% higher than its parent strain.Generation transmission experiment had proved the mutant strain had genetic stability and can be used as experiment strain for further uses.
