摘要
目的:观察IL-23对溃疡性结肠炎(UC)患者、感染性腹泻及健康对照者外周血单个核细胞(PBMCs)分泌IL-17的影响。方法:分离并培养17例溃疡性结肠炎患者、10例感染性腹泻、13例健康对照者外周血单个核细胞;ELISA方法检测IL-23刺激前后各组PBMCs上清液中IL-17的水平。结果:未经IL-23刺激的各组PBMCs上清液中IL-17的水平分别为(29.492±6.631)pg/ml、(18.183±3.110)pg/ml、(16.848±3.903)pg/ml,其中UC组明显高于另两组(P<0.05);IL-23可促进各组PBMCs分泌IL-17,且UC组(63.631±11.793)pg/ml较感染性腹泻组(27.868±8.314)pg/ml及健康对照组(26.533±6.422)pg/ml升高更显著(P<0.05)。结论:IL-23可能通过诱导IL-17的分泌而使后者在UC的慢性病程中发挥重要作用,IL-23/IL-17免疫通路失调可能破坏机体免疫耐受而导致UC疾病的发生。
Objective:To observe the effect of IL-23 on secretion of IL- 17 in the peripheral blood mononuclear cells(PBMCs) of ulcerative colitis patients(UC), Infectious diarrhea patients and healthy controls.Methods:PBMCs(peripheral blood mononuelear cells), isolated from 17 eases of UC, 10 eases of Infectious diarrhea patients and 13 healthy controls;The amounts of IL- 17 induced or not by IL-23 in supematants of cultured PBMCs from each group were measured by ELISA.Results:The amounts of IL-17 which were not induced by IL-23 in each group were (29.492±6.631 )pg/ml, ( 18.183±3.110)pg/ml, ( 16.848±3.903)pg/ml respectively, UC group were higher than other groups(P〈0.05);IL-23 might stimulate the production of IL-17 of the PBMCs from all groups,and that of UC group was significantly higher than that of other two groups(P〈0.05);Conclusion:IL-23 might play a critical role in the development of UC through inducing production of IL-17.Impaired IL-23/IL-17 path way may induce UC by destructing immune tolerance.
