新型Hsp90抑制剂-SNX-2112的小鼠骨髓微核试验研究

摘要 目的:通过小鼠骨髓微核试验来研究SNX-2112的遗传毒性。方法:昆明小白鼠尾静脉注射SNX-2112,24 h后处死动物,取双侧股骨制备骨髓细胞悬液,进行骨髓推片和姬姆萨染色,在显微镜下选择细胞分布均匀和着色好的区域阅片,计数含微核嗜多染红细胞(polychromatic erythrocyte,PCE)数。比较各实验组及阳性对照组与阴性对照组的微核率。结果:空白溶媒组及SNX-2112低剂量组与阴性对照相比差别均无显著性(P>0.05),SNX-2112中剂量及SNX-2112高剂量组与阴性对照相比差别有统计意义(P(0.05)。结论:SNX-2112在一定剂量范围内对小鼠染色体无明显的损伤作用。 Objective:By mouse bone marrow micronucleus test to study the genotoxicity of SNX-2112.Methods:The KM mice were sacrificed at 24 h after caudal vein injection of SNX-2112.Bone marrow extracted from bilateral femur was used to make cell suspension.Slide specimens were fixed,dried,and dipped into Giemsa.The district which had complete cell,well-distributed,and well-coloration,was selected and counted under microscope.Comparing the experimental group and the positive control group and negative control.Results:Blank solvent group and SNX-2112 low-dose group,compared with the negative control was no significant difference(P﹥0.05).SNX-2112 middle-dose and SNX-2112 high-dose,compared with the negative control was significant difference(P﹤0.05).Conclusion:In a certain dose range,SNX-2112 has no obvious damage on mouse chromosome.
