注射金丝桃素并525nm激光照射对鲜红斑痣模型罗曼鸡鸡冠的血管损伤作用 被引量:2

摘要 目的探讨注射金丝桃素(hypericin,HY)并525nm激光照射对鲜红斑痣模型罗曼鸡鸡冠的作用。方法取6月龄雄性罗曼鸡36只,按随机数字表法分为6组:正常对照组、激光对照组、血卟啉1mg/kg阳性对照组、HY0.3mg/kg组、HY1mg/kg组、HY3mg/kg组,每组6只。除正常对照组外,对各组鸡冠单侧进行激光照射,未处理一侧作为自身对照。罗曼鸡静脉注射金丝桃素后,立即用激光照射鸡冠直径2cm的区域,鸡冠剂量为功率密度100mW/cm2。各组(除正常对照组)给药并525nm激光照射后每天观察照相,处理7、14d后处死动物,取鸡冠进行病理检查,并对鸡冠毛细血管计数。结果HY给药组和血卟啉对照组从外观上可对鸡冠表皮产生不同程度的损伤,造成明显水肿、结痂、坏死。HY1mg/kg,HY3mg/kg组及血卟啉1mg/kg阳性对照组对鸡冠的表皮损伤尤其明显,产生明显的坏死、结痂。HE组织学观察显示,HY各给药组及血卟啉对照组能导致鸡冠表皮增生,减少真皮毛细血管数,其毛细血管减少率与正常对照组及激光对照组相比差异有统计学意义(P<0.01),激光对照组与正常对照组相比差异不大。结论HY静脉注射给药并525nm激光照射对鸡冠真皮毛细血管有较好的破坏作用,能对鲜红斑痣模型鸡冠起到明显损伤作用。 Objective To explore the effect of hypericin (HY) combined with 525 nm photodynamic therapy (PDT) on vascular lesion of cock comb as the model of port wine stains.Methods Totally 36 Longman cocks at the age of 6 months were divided into 6 groups randomly,including control group,irradiation group,positive control group (hemoporphyrin 1 mg/kg),0.3 mg/kg HY group,1 mg/kg HY group,and 3 mg/kg HY group.Aspots of 2.0 cm in diameter on one side of cock comb was irradiated by 525 nm laser at a power density of 100 mW/cm2 immediately after corresponding photosensitiser intravenous injection.While the other side of cock comb served as control and received no irradiation.Visual observation and photos were recorded every day,pathological changes were observed and the reduction rate of comb’s capillary of all groups was counted in 7 and 14 d after photodynamic therapy.Results HY and 1 mg/kg hemoporphyrin damaged the epidermis,such as edema,necrosis and incrustation,and abated the red color of cock bomb obviously,especially in groups of 1 and 3 mg/kg HY combined with 1 mg/kg hemoporphyrin injection.At the same time,epidermis hyperplasia,and the reduction rate of comb’s capillary were decreased by HY and hemoporphyrin with 525 nm photodynamic therapy.Control group and irradiation group had no such changes (P0.05).Conclusion Intravenous injection of HY associated with photodynamic irradiation at 525 nm is proved to have effective damage to cutaneous micrangium of cock comb
