摘要
目的建立一种酶联免疫吸附试验(ELISA)方法 ,测定比格犬血液中重组水蛭素浓度,进而研究其药动学特点。方法选取合格比格犬6只,静脉注射重组水蛭素0.2 mg/kg,于给药后2、5、10、20、40、60、120、240和480 min取血2 mL,分离血清,用ELISA检测其血药浓度。结果静脉注射重组水蛭素的T1/2α(0.37±0.029)h,T1/2β(2.64±0.92)h;ATU0-T(372.98±27.87)ng/(mL.h)。结论经ELISA测定血中的药物浓度,药时曲线经3P97拟合,表明静脉注射重组水蛭素在动物体内为二室模型。
Objective To develop a ELISA method for determining r-hirudin(rH) in Beagle dog's plasma so as to provide a methodology for the study of its harmacokineties. Methods 6 eligible beagle clogs were intravenously injected with rH(0.2mg/kg). The method of ELISA was used to detect the rH in Beagle dog's plasma after iv administration 2,5,10,20,40,60,120,240 and 480 min. Results The pharmacokinetie parameters determined by ELISA method were as follows:T1/2a were (0.37±0. 029)h,T1/2βwere (2.64±0.92)h,ATU0-T were (372.98±27.87)ng/ (mE ·h). Conclusion The result showed that drug concentration-time curve was conformed to two compartment open models.
