摘要
类钙调磷酸酶B蛋白(calcineurin B-like proteins,CBLs)是植物中1类重要的Ca2+结合蛋白,由其所介导的Ca2+信号途径广泛参与植物对多种非生物胁迫和某些生长发育信号的应答反应。本论文以水稻OsCBL6序列在GenBank数据库进行TBLASTN查询,获得其在玉米中推测的同源基因ZmCBL6的全长cDNA序列,然后用RT-PCR方法对ZmCBL6基因的编码框cDNA进行了克隆,用生物信息学方法对其编码蛋白的功能域、系统进化和亚细胞定位等进行了分析,并构建了其植物表达载体,为进一步研究其亚细胞定位和通过转基因植物等验证其功能奠定了基础。
The calcineurin B-like proteins(CBLs)is an important family of calcium-binding proteins in plants,and the calcium signal pathways mediated by CBLs extensively participate in the responses to a variety of abiotic stresses and some developmental cues in plants.In this paper,a full-length cDNA sequence of ZmCBL6 which is a putative homologous gene of rice(Oryza sativa) OsCBL6 in maize was obtained by searching the GenBank database using OsCBL6 amino acid sequence as the direct query with TBLASTN program.The complete coding region cDNA of ZmCBL6 was amplified by reverse transcription polymerase chain reaction(RT-PCR),and the domains,the phylogenetic relationship and the subcellular localization of the predicted ZmCBL6 protein were analyzed using bioinformatic methods.Moreover,the plant expression vector for the gene was constructed.The work laid a foundation for the analyses of the subcellular localization of ZmCBL6 protein and its function in transgenic plants in the future.
