液相组合化学中分离纯化方法的研究与进展

摘要 随着高通量有机合成技术的不断提高,合成的化合物库日益增大,使用传统的柱色谱等手段难以满足分离速度的需求;同时由于低纯度的化合物在活性筛选过程中往往产生不真实的数据,干扰实验结果,所以纯化后化合物库的纯度标准需要提高。为解决液相组合化学合成产物的分离速度和质量问题,组合化学家们相继发展了制备色谱和氟试剂等方法和试剂。综述了各种液相组合合成分离方法及特点,重点介绍了近年发展起来的氟化学分离技术。 High throughput organic synthesis has produced huge compound libraries,which greatly exceed the production capability of traditional separation technologies.Meanwhile,compounds with low purity often give false data in biological screening.The purity standards for the 'purified' products must be upgraded.To get better products much more quickly,combinational chemists have developed appropriate techniques and reagents,such as preparative chromatography and fluorinating technology.In this paper,the advantages and disadvantages of currently used separation methods were evaluated and the fluorine chemistry separation technique was introduced in detail.
