头针对脑缺血再灌注大鼠脑源性神经营养因子表达的影响 被引量:8

摘要 目的探讨头针对脑缺血再灌注模型大鼠脑源性神经营养因子(BDNF)表达的影响,探讨头针治疗脑缺血的可能机制。方法将70只健康Wistar雌性大鼠随机分为假手术组、模型组、头针组(模型组和头针组再根据缺血再灌注时间的不同,各随机分为7、14、28d3个亚组)。采用大脑中动脉线栓法制备大脑中动脉闭塞(MCAO)再灌注模型,选取顶颞后斜线,顶颞前斜线,进针后接韩氏穴位神经刺激仪,取疏密波,频率2Hz/100Hz,强度2mA,每次20min,每日1次。应用神经功能缺损评分(NSS)、免疫组织化学法观察脑缺血再灌注各时间点头针对模型大鼠神经功能缺损、缺血区海马齿状回(DG)BDNF含量的影响。结果头针组与模型组各时相上的组间比较,(1)NSS指标:第28日头针组的NSS显著低于模型组(P<0.05)。(2)缺血区海马齿状回BDNF表达情况:假手术组与头针组和模型组比较有显著差异,头针组与模型组各时间相上组间比较,缺血再灌注7d时头针组与模型组差异有统计学意义。结论头针可通过增加内源性BDNF的含量以促进急性脑缺血再灌注损伤过程中神经干细胞的修复,实现对缺血脑组织保护的目的。 Objective:To observe scalp-acupuncture on BDNF expression in cerebral ischemia-reperfusion rats and to explore the possible mechanisms of scalp-acupuncture in this disease.Methods:70 rats were divided randomly into the sham-operation group,the model group and the scalp-acupuncture group.According to the differences of the ischemia-reperfusion time points,the model group and the scalp-acupuncture group were divided randomly into the 7d group,the 14d group and the 28d group.After middle cerebral artery occlusion(MCAO)establishment,the anterior and the posterior of oblique line of vertex-temporal were inserted by the Han's Electro-Acupuncture apparatus.It was alternated the frequency of 2 Hz and 100 Hz at an intensity level of 2 mA.The treatment frequency was 20min per time and one time each day.After needle,the neurologic impairment and the BDNF of DG in ischemia area were applied by the NSS and the immunohistochemistry method.Results:Compared every time points with the scalp-acupuncture group and the model group,there were some indices including the NSS and the BDNF expression of DG in ischemia area.The NSS of the 28d group was less significantly than that of the model group.There was significant difference in the BDNF expression between the sham-operation group and the model group.Meanwhile,this variance was also remarkable between the 7d group and the model group.Conclusion:Scalp-acupuncture is beneficial for neurofunctional rehabilitation by promoting the rehabilitation of the nerve cell which is through increasing the BDNF content.
