头针对急性脑缺血再灌注大鼠细胞凋亡的影响 被引量:10

摘要 目的探讨头针治疗脑缺血的可能机制。方法将90只健康SD雌性大鼠随机分为假手术组、模型组、头针组模型组和头针组再根据缺血再灌注时间的不同而随机分为6、24、48、72h共八个亚组,采用大脑中动脉线栓法制备大脑中动脉闭塞(MCAO)再灌注模型,选取顶颞后斜线,顶颞前斜线、进针后接韩氏穴位神经刺激仪治疗;观察各时间点大鼠神经功能缺损、脑神经元TUNEL阳性细胞数的变化。结果头针组与模型组各时相上的组间比较,第72小时头针组的NSS显著低于模型组;模型组缺血侧神经细胞凋亡数随再灌注时间延长而增加,24h达高峰;头针治疗可明显减少神经细胞凋亡数,以24、48h显著。结论头针有利于大鼠脑神经功能的恢复,可减轻急性脑缺血再灌注后细胞凋亡,并在一定范围内降低阳性凋亡细胞的表达。头针抗脑缺血的作用机制可能为抑制细胞凋亡反应,减轻脑缺血再灌注损伤,实现脑组织的保护作用。 Objective: To explore the possible mechanisms of scalp- acupuncture (SA) in treating cerebral ischemia. Methods: Rat models with middle cerebral artery occlusion (MCAO) were used and 48 models were randomized into model group and SA group(inserting needles along anterior oblique line of vertextemporal and posterior oblique line of vertex -temporal, soon after when a Han's Point -nerve Stimulating Apparatus was used) and we observed changes of neurofunctional defect, TUNEL positive changes in cells of neurons in 6, 24, 48 and 72h. Results: On NSS at each phase point: SA group demonstrated obvious decline in NSS at 72h when compared with that of model group. The positive rate of apoptosis: model ischemic neuronal cell apoptosis increase with a few extension of reper fusion time, reached the peak in 24h, scalp - acupuncture treatment could significantly reduce the number of neuronal apoptosis, especially in 24h and 4gh sub - groups. Conclusion: Scalp - acupuncture is beneficial for neurofunctional rehabilitation, which can suppress cell apoptosis, within a certain range. The positive apoptotic expression, in acute ischemia reperfusion injury. The possible mechanisms of SA against brain ischemia may be concerned with the effectiveness of inhibiting cell Apoptosis reaction, so the cerebral ischemia reperfusion injury can be attenuated by the regulation for the disordered immune state.
