摘要
目的:探讨黄芩茎叶总黄酮(Scutellaria Baicalensis stem-leaf Total Flavonoid,SSTF)对大鼠心肌缺血再灌注损伤的保护作用及其机制。方法:40只SD大鼠随机分为假手术组、缺血再灌注组、SSTF组。SSTF组大鼠于术前1周灌胃不同剂量的SSTF(17.5mg/kg/d、35mg/kg/d、70mg/kg/d),模型组及假手术组给予等体积生理盐水。采用结扎大鼠左冠状动脉前降支的方法制备心肌缺血再灌注(myocardial ischemia reperfusion injury,MIRI)模型,缺血30min再灌注2h,应用流式细胞仪观察心肌细胞凋亡情况,同时检测血清乳酸脱氢酶(lactate dehydrogenase,LDH)活性、心肌组织超氧化物歧化酶(superoxide dismutase,SOD)活性及丙二醛(malondialdehydebis,MDA)含量。结果:SSTF可以抑制损伤大鼠心肌细胞凋亡的发生,并显著降低MDA含量,使乳酸脱氢酶释放减少,提高SOD的活性,上述指标与缺血再灌注组相比均有显著性差异。结论:SSTF对大鼠心肌缺血再灌注损伤具有保护作用,其机制可能与清除氧自由基、增强抗氧化酶活性、抑制心肌细胞凋亡有关。
Objective:To explore the protective effects and mechanism about SSTF against myocardial ischemia reperfusion injury in Rats.Methods:40 SD rats were divided into sham operation group,Ischemia Reperfusion group and SSTF group at random.Rats in SSTF group were Perfused with SSTF a week before operation,and to the sham operation group and the model group,saline was given instead.Myocardial Ischemia reperfusion injury model was made by ligating the left descend coronary artery 30min and reperfusing 2 hours.After operation,the myocardial cell apotosis by Flow cytometer(FCM)was observed;The content of MDA and activity of LDH and SOD in myocardium were measured.Results:Compared with Ischemia Reperfusion group,SSTF can inhibit the myocardial cell apotosis,decrease the content of MDA and the activity of LDH,increase the activity of SOD.Conclusions:SSTF can protect myocardium damage induced by IRI.The mechanism of cardioprotection may be associated with clearing oxygen free radial(OFR),increasing the activity of SOD,inhibiting apoptosis of myocardium.
